Cubic phase Containing Hydrophobically Modified Glucose Oxidase and Acidic Proteinoid for Glucose-sensitive Release

Glucose-sensitive monoolein (MO) cubic phase is prepared by containing hydrophobically modified glucose oxdiase (HmGOD) and acidic proteinoid for a pH-dependent and glucose-sensitive system. GOD was hydrophobically modified by palmitic acid N-hydroxysuccinimide ester. And poly (glutamic acid-co-leucine) prepared by a melt-condensation method is used as acidic proteinoid and included in water chnnel of cubic phase. When pH is lower than pI of acidic proteinoid, the acidic proteinoid was condensed, promoting the release of insulin loaded in the cubic phase. High concentration of glucose also promotes the release of insulin from cubic phase. That’s because HmGOD immobilized in the lipid matrix of cubic phase interacts with glucose, reducing pH in water channel in cubic phase, which further affects the condensation of acidic proteinoid and increases the release of insulin. Therefore the developed cubic phase could control the release of insulin depending on pH change and glucose concentration.
